Mass Spectrometry-Based Proteomic and Immunoproteomic Analyses of the Candida albicans Hyphal Secretome Reveal Diagnostic Biomarker Candidates for Invasive Candidiasis

چکیده

Invasive candidiasis (IC) is associated with high morbidity and mortality in hospitalized patients if not diagnosed early. Long-term use of central venous catheters a predisposing factor for IC. Hyphal forms Candida albicans (the major etiological agent IC) are related to invasion host tissues. The secreted proteins hyphae involved virulence, interaction, immune response, evasion. To identify IC diagnostic biomarker candidates, we characterized the C. hyphal secretome by gel-free proteomic analysis, further assessed antibody-reactivity patterns this subproteome serum pools from 12 non-catheter-associated (ncIC), 11 catheter-associated (cIC), non-IC patients. We identified 301 stratified stem extracellular region, cell wall, surface, or intracellular compartments. ncIC cIC had higher antibody levels than Seven were be immunogenic (Bgl2, Eno1, Pgk1, Glx3, Sap5, Pra1 Tdh3). Antibody-reactivity Bgl2, Pgk1 Glx3 discriminated patients, while those Tdh3 differentiated between These may useful development future tests.

Mass spectrometric analysis of the secretome of Candida albicans.

The pathogenic fungus Candida albicans secretes a considerable number of hydrolases and other proteins. In-depth studies of the C. albicans secretome could thus provide new candidates for diagnostic markers and vaccine development. We compared various growth conditions differing in pH, temperature and the presence of the hyphal inducer N-acetylglucosamine. The polypeptide content of the growth ...

Biomarker-guided antifungal stewardship policies for patients with invasive candidiasis

Invasive fungal infections (IFIs) are among the life-threatening issues in patients with impaired immune system. High administration of antifungals in these patients imposes a heavy economic burden on the national health system. In addition, despite the usually expensive antifungal regimens, the mortality rate due to fungal infections is still high, resulting in the loss of hundreds of lives pe...

Pharmacoeconomic analysis of antifungal therapy for primary treatment of invasive candidiasis caused by Candida albicans and non-albicans Candida species

BACKGROUND Cost-effectiveness studies of echinocandins for the treatment of invasive candidiasis, including candidemia, are rare in Asia. No study has determined whether echinocandins are cost-effective for both Candida albicans and non-albicans Candida species. There have been no economic evaluations that compare non-echinocandins with the three available echinocandins. This study was aimed to...

Mass spectrometry-based proteomic analyses of contact lens deposition

PURPOSE The purpose of this report is to describe the contact lens deposition proteome associated with two silicone hydrogel contact lenses and care solutions using a mass spectrometric-based approach. METHODS This was a randomized, controlled, examiner-masked crossover clinical trial that included 48 participants. Lenses and no-rub care solutions evaluated included galyfilcon A (Acuvue Advan...

Rapid Species Diagnosis for Invasive Candidiasis Using Mass Spectrometry

BACKGROUND Matrix-assisted laser desorption ionisation time of flight mass spectrometry (MALDI TOF-MS) allows the identification of most bacteria and an increasing number of fungi. The potential for the highest clinical benefit of such methods would be in severe acute infections that require prompt treatment adapted to the infecting species. Our objective was to determine whether yeasts could b...
